In this study, using Chlamydomonas as a material, we investigated signaling molecules such as A-kinase (PKA), G-kinase (PKG), and C-kinase (PKC) that are widely present in animal cells but not recognized in higher plants. A cDNA fragment (designated as PK1) was amplified from Chlamydomonas by RT-PCR using a set of oligonucleotide primers, corresponding to the conserved amino acid sequences of animal-type AGC kinases. A Chlamydomonas cDNA library was screened with the PCR-amplified fragment as a probe, we identified a partial sequence of PKA -like protein kinase gene (designated as partial CPK1). Furthermore, two open-reading frames of the protein kinase genes were obtained by the database analysis of Chlamydomonas EST libraries. Nucleotide sequence of each protein kinase has 100% or 66% similarities with the partial CPK1, respectively. These two protein kinase genes designated CPK1 and CPK2 , encode both regulatory domains of PKG in their N terminal region and catalytic domains of PKG in their C terminus. CPK1 gene was expressed in vegetative cells of Chlamydomonas , whereas, CPK2 gene was expressed in gamete cells. FLAG-tagged CPK1 expressed in E.coli were harvested by immunoprecipitation using anti-FLAG antibody. The immunoprecipitates reacted with three kinds of protein kinase substrates under cAMP or cGMP. As a result, CPK1 phosphorylates most strongly the PKG specific substrate (BPDE tide) under the presence of cGMP. These results suggest that CPK1 is a typical PKG and functions in vegetative cells of Chlamydomonas .
